Purpose
The purpose of the core facility is to provide access to a high content screening microscope. This enables automated imaging and analysis of a large number of cells in a multi-well format for quantitative microscopy.
Instrument
The HCA Core Facility ensures the use of a Thermo Scientific™ CellInsight™ CX5 High Content Screening (HCS) Platform.
Camera: Photometrics™ X1 CCD high-resolution fluorescence camera
- High-resolution, thermoelectrically cooled to 0°C
- 54 micron pixel size for high resolution detail in image capture and large field of view 2,208 by 2,208 pixel array
Light source:
The CX5 platform uses high-output LED light and is configured for imaging in the UV range through near-IR range with the provided filter sets. The platform offers the following excitation/ emission capabilities with Invitrogen™ Alexa Fluor™ dyes:
- 386/440 (e.g., Alexa Fluor 350 dye)
- 485/521 (e.g., Alexa Fluor 488 dye)
- 560/607 (e.g., Alexa Fluor 594 dye)
- 650/694 (e.g., Alexa Fluor 647 dye)
- 740/810 (e.g., Alexa Fluor 750 dye)
Objectives: Olympus™ 10x (0.3 NA) and 20x (0.4 NA) objectives
Detection Method: Brightfield, Fluorescent
Format: The system is compatible with most optically clear, flat, 8-well chamber slides; 6-, 12-, 24-, 48-, 96-, 384-, and 1,536-well microplates.
Software: Thermo Scientific™ HCS Studio 5.0 Cell Analysis Software for image acquisition and data analysis.
Equipped with an Onstage Incubator:
It enables precise control of temperature, humidity, and CO2 for time-lapse imaging of live cells under both physiological and non-physiological conditions. Environmental settings and image acquisition parameters are all seamlessly integrated into the HCS Studio software creating a powerful, walk-away platform for live cell quantitative imaging.
Advantages of the Onstage Incubator:
- Fully integrated environmental chamber for live cell time-lapse imaging
- Precisely maintain physiological or non-physiological conditions
- Simple scheduling to define frequency of imaging over a time course
- Powerful kinetic and motility measurements that increase information gathered from each cell over time
