El\u0151ad\u00e1suk kivonatait al\u00e1bb olvashatj\u00e1k:<\/p>\n<div class=\"exp-col-content-holder\"><a class=\"expand-cnt-link\" href=\"#\">Sz\u0151cs Em\u0151ke: A bursa Fabriciiben zajl\u00f3 lymphoid follikulogenezis karakteriz\u00e1l\u00e1sa<\/a><\/p>\n<div class=\"hidden-content\">\n<p class=\"hiddenContentp\" style=\"text-align: justify\">A bursa Fabricii (BF) a madarak b\u00e9lhez-asszoci\u00e1lt primer nyirokszerve, a B-lymphocyt\u00e1k prolifer\u00e1ci\u00f3j\u00e1nak, differenci\u00e1l\u00f3d\u00e1s\u00e1nak, az immunreperto\u00e1r kialakul\u00e1s\u00e1nak helye. Az embryon\u00e1lis fejl\u0151d\u00e9s kezdet\u00e9n a BF epithelialis telepe a cloaca diszt\u00e1lis, ektoderma-eredet\u0171 szakasz\u00e1b\u00f3l indul ki. Az organogenezis idej\u00e9n a BF h\u00e1mkezdem\u00e9ny\u00e9t vastag mesenchym\u00e1lis sejtr\u00e9teg bor\u00edtja, amelyet v\u00e9r eredet\u0171 \u0151ssejtek (B-lymphocyta, makrof\u00e1g \u00e9s dendritikus prekurzorok) n\u00e9pes\u00edtenek be. A v\u00e9reredet\u0171 myeloid prekurzorok a mesenchym\u00e1ban a h\u00e1msejteket follikulusbimb\u00f3k k\u00e9pz\u00e9s\u00e9re k\u00e9sztetik. A B-lymphocyta prekurzorok tov\u00e1bbi differenci\u00e1l\u00f3d\u00e1s\u00e1t a BF follikulusokat alkot\u00f3 dendritikus-, makrof\u00e1g-, \u00e9s h\u00e1mretikulum sejtekb\u0151l \u00e1ll\u00f3 mikrok\u00f6rnyezet hat\u00e1rozza meg. A strom\u00e1lis mikrok\u00f6rnyezet jelent\u0151s\u00e9ge a lymphopoiezisben f\u0151leg az \u0151ssejtek differenci\u00e1l\u00f3d\u00e1s\u00e1t ir\u00e1ny\u00edt\u00f3 n\u00f6veked\u00e9si faktorok \u00e9s citokinek felfedez\u00e9s\u00e9vel \u00e9rt\u00e9kel\u0151d\u00f6tt fel. Jelen munk\u00e1nk sor\u00e1n hisztol\u00f3giai \u00e9s immuncitok\u00e9miai m\u00f3dszerrel tanulm\u00e1nyoztuk a myeloid sejtek megjelen\u00e9s\u00e9t \u00e9s differenci\u00e1l\u00f3d\u00e1s\u00e1t, amelyek elind\u00edtj\u00e1k a BF lymphoid follikulusainak ontogenezis\u00e9t a B-sejtek bejut\u00e1sa el\u0151tt. A 9 napos csirke embryoban az EIVE12 monoklon\u00e1lis ellenanyag (mAb) a h\u00e1m alatti mesenchym\u00e1ban kis csoportokat k\u00e9pez\u0151 kerek sejteket jel\u00f6l, melyek h\u00e1mba v\u00e1ndorl\u00e1s\u00e1t k\u00f6veti a follikulusok kezdem\u00e9ny\u00e9t alkot\u00f3 h\u00e1mbimb\u00f3k megjelen\u00e9se. Egy nappal k\u00e9s\u0151bb, CSF1R+, illetve TIM4 receptorokat expressz\u00e1l\u00f3 ny\u00falv\u00e1nyos sejtek koloniz\u00e1lj\u00e1k a h\u00e1mbimb\u00f3kat, amiben 24-48 \u00f3r\u00e1val k\u00e9s\u0151bb CXCR4+\/chB6+\/IgM+ B-sejt prekurzorok telepednek meg. Az embryon\u00e1lis fejl\u0151d\u00e9s el\u0151rehaladt\u00e1val a CSF1R+, illetve TIM4+ sejtek sz\u00e1ma megn\u0151, \u00e9s a 16-18 napos embri\u00f3 follikulusaiban m\u00e1r dendritikus, valamint makrof\u00e1g morfol\u00f3gi\u00e1t mutatnak. Kikel\u00e9st k\u00f6vet\u0151en a BF-ben k\u00e9pz\u0151d\u0151 chB6+ B-lymphocyt\u00e1k a perif\u00e9ri\u00e1ra v\u00e1ndorolva a szekunder nyirokszerveket koloniz\u00e1lj\u00e1k. K\u00f6vetkeztet\u00e9s: Az EIVE12 mAb \u00e1ltal felismert sejt epitheli\u00e1lis inv\u00e1zi\u00f3ja az embryon\u00e1lis \u00e9let sor\u00e1n j\u00f3val megel\u0151zi a dendritikus\/makrof\u00e1g sejtek follikulusbimb\u00f3kba t\u00f6rt\u00e9n\u0151 bev\u00e1ndorl\u00e1s\u00e1t. Ez az \u00faj megfigyel\u00e9s arra utal, hogy az eddigi elk\u00e9pzel\u00e9ssel szemben nem a dendritikus prekurzor sejtek \u00e9s a h\u00e1m dendro-epitheli\u00e1lis k\u00f6lcs\u00f6nhat\u00e1sa, hanem egy \u00faj sejtt\u00edpus, az EIVE12+ \u201elymphoid follikulusbimb\u00f3 induk\u00e1l\u00f3\u201d sejtek inv\u00e1zi\u00f3ja ind\u00edtja el a follikulogenezist.<\/p>\n<\/div>\n<\/div>\n<div class=\"exp-col-content-holder\"><a class=\"expand-cnt-link\" href=\"#\">So\u00f3s \u00c1d\u00e1m: A k\u00e9reg\u00e1llom\u00e1ny sejtes \u00e9s molekul\u00e1ris \u00f6sszet\u00e9tele a bursa Fabricii limfoid follikulusaiban<\/a><\/p>\n<div class=\"hidden-content\">\n<p class=\"hiddenContentp\" style=\"text-align: justify\">A bursa Fabricii a madarak primer nyirokszerve, amelynek szerkezeti \u00e9s m\u0171k\u00f6d\u00e9si alapegys\u00e9geiben, a limfoid follikulusokban zajlik a B-sejtek prolifer\u00e1ci\u00f3ja \u00e9s differenci\u00e1l\u00f3d\u00e1sa. A follikulusok k\u00e9t, sz\u00f6vettanilag \u00e9s fejl\u0151d\u00e9stanilag j\u00f3l elk\u00fcl\u00f6n\u00fclt kompartmentb\u0151l, az ektoderm\u00e1lis eredet\u0171 vel\u0151\u00e1llom\u00e1nyb\u00f3l \u00e9s a mezoderm\u00e1lis eredet\u0171 k\u00e9reg\u00e1llom\u00e1nyb\u00f3l \u00e1llnak. Annak ellen\u00e9re, hogy a B-sejtek \u00e9r\u00e9shez sz\u00fcks\u00e9ges follikul\u00e1ris vel\u0151\u00e1llom\u00e1ny sz\u00f6vettana r\u00e9szletesen karakteriz\u00e1lt, az ontogenetikailag k\u00e9s\u0151bb megjelen\u0151 kortik\u00e1lis r\u00e9gi\u00f3 sz\u00f6veti \u00e9s molekul\u00e1ris \u00f6sszet\u00e9tele nem ismert. Kutat\u00e1sunk sor\u00e1n a bursa Fabricii follikulusok k\u00e9reg\u00e1llom\u00e1ny\u00e1nak sejtsz\u00ednt\u0171 \u00e9s molekul\u00e1ris elemz\u00e9s\u00e9t v\u00e9gezt\u00fck immuncitok\u00e9miai \u00e9s hisztol\u00f3giai m\u00f3dszerekkel, kieg\u00e9sz\u00edtve sejtteny\u00e9szt\u00e9ses \u00e9s embri\u00f3manipul\u00e1ci\u00f3s technik\u00e1kkal. Az IgM+ molekul\u00e1kat kifejez\u0151 medull\u00e1ris B-sejtekkel ellent\u00e9tben a k\u00e9reg B-sejtjei CXCR4 pozit\u00edvak, \u00e9s IgM-et alacsonyan expressz\u00e1lnak. A follikul\u00e1ris k\u00e9reg\u00e1llom\u00e1nyban dendritikus sejt nem tal\u00e1lhat\u00f3, viszont CSF1R+\/TIM4+\/Lep100+ makrof\u00e1gok egyenletesen el\u0151fordulnak. A limfo-mieloid sejtekben gazdag kortex alapv\u00e1z\u00e1t dezmint \u00e9s vimentint kifejez\u0151 k\u00e9rgi retikulumsejtek alkotj\u00e1k, amelyek kollag\u00e9n (I-es t\u00edpus\u00fa, III-, IV-, VI t\u00edpus\u00fa), glikoprotein (laminin, fibronektin, fibrillin, tenaszcin-C), illetve proteoglik\u00e1nokban gazdag ECM h\u00e1l\u00f3zatot termelnek. Az ECM r\u00e9szletes karakteriz\u00e1l\u00e1s sor\u00e1n azt tal\u00e1ltuk, hogy m\u00edg a m\u00e1trix feh\u00e9rj\u00e9k t\u00f6bbs\u00e9ge korai embrion\u00e1lis burs\u00e1ban is kifejez\u0151dik, addig a tenaszcin-C expresszi\u00f3ja el\u0151sz\u00f6r csak a kikel\u00e9s t\u00e1j\u00e1n, a 16-18 napos embri\u00f3 follikuluskezdem\u00e9nyei k\u00f6r\u00fcl figyelhet\u0151 meg. A kikel\u00e9st k\u00f6vet\u0151en a k\u00e9reg\u00e1llom\u00e1nyra specifikus tenaszcin-C, jellegzetes eloszl\u00e1st mutatva a kapill\u00e1risok k\u00f6r\u00fcl koncentr\u00e1l\u00f3dik. A tenaszcin-C funkci\u00f3j\u00e1nak in vitro vizsg\u00e1lata igazolta, hogy a B-sejtek migr\u00e1ci\u00f3ja szempontj\u00e1b\u00f3l g\u00e1tl\u00f3 k\u00f6rnyezetet k\u00e9pvisel. In vivo RCAS-Shh retrov\u00edrus vektor \u00e1ltal el\u0151id\u00e9zett tenaszcin-C t\u00falexpressz\u00e1ltat\u00e1s is g\u00e1tolja a fejl\u0151d\u0151 follikulusok B-sejtes koloniz\u00e1ci\u00f3j\u00e1t. Eredm\u00e9nyeinket \u00f6sszefoglalva kijelenthetj\u00fck hogy 1.) a k\u00e9reg\u00e1llom\u00e1ny fejl\u0151d\u00e9se a kikel\u00e9s el\u0151tt 5 nappal megkezd\u0151dik. 2) a k\u00e9reg\u00e1llom\u00e1ny B-sejt popul\u00e1ci\u00f3ja Bu- 1+\/CXCR4+\/IgM low + expresszi\u00f3s mint\u00e1zatot mutat \u00e9s CSF1R+ dendritikus sejteket nem tartalmaz, viszont rendelkezik egy CSF1R+\/TIM4+\/Lep100+ makrof\u00e1g popul\u00e1ci\u00f3val. 3) a k\u00e9reg\u00e1llom\u00e1ny v\u00e1zrendszer\u00e9t mezenchim\u00e1lis retikulum sejtek k\u00e9pezik, amelyek tenaszcin-C- ben gazdag extracellul\u00e1ris m\u00e1trixot termelnek. 4) In vivo \u00e9s in vitro k\u00eds\u00e9rletek szerint a tenaszcin-C g\u00e1tl\u00f3 k\u00f6rnyezetet jelent a v\u00e1ndorl\u00f3 B-sejtek
