A vegy\u00fcletcsoportot Knoll \u00e9s mtsai. \u00edrt\u00e1k le (1999); a vegy\u00fcletcsoport el\u0151\u00e1ll\u00edt\u00e1sa a (-)-deprenyl (selegilin) szerkezet\u00e9b\u0151l indult ki. Az \u00fajabb vegy\u00fcletek, a (-)-1-(benzofuran-2-yl)-2-propilamino-pent\u00e1n (BPAP) \u00e9s sz\u00e1rmaz\u00e9kai (PPAP, IPAP) azonban (-)-deprenylt\u0151l elt\u00e9r\u0151en, nagyobb d\u00f3zisban sem rendelkeznek B t\u00edpus\u00fa monoamin oxid\u00e1z (MAO-B) g\u00e1tl\u00f3, ill. katecholamin felszabadul\u00e1st serkent\u0151 hat\u00e1ssal. Az enhancer hat\u00e1s\u00fa vegy\u00fcletek a biog\u00e9n aminok neurok\u00e9miai transzmisszi\u00f3j\u00e1t elt\u00e9r\u0151 er\u0151ss\u00e9ggel befoly\u00e1solhatj\u00e1k, hat\u00e1sukat els\u0151sorban a stimulus induk\u00e1lta biog\u00e9n amin felszabadul\u00e1sra fejtik ki. Az enhancer hat\u00e1s a fentiek alapj\u00e1n f\u00fcggetlen az amfetamin vegy\u00fcletekre jellemz\u0151 biog\u00e9n amin felszabadul\u00e1st fokoz\u00f3 hat\u00e1st\u00f3l; f\u00fcggetlen tov\u00e1bb\u00e1 a MAO-B g\u00e1tl\u00f3 hat\u00e1st\u00f3l, hiszen az ugyancsak MAO-B g\u00e1tl\u00f3 rasagilin enhancer hat\u00e1ssal nem \u00a0rendelkezik.<\/p>\n<p>Az enhancer hat\u00e1s molekul\u00e1ris szint\u0171 mechanizmusa eddig nem ismert pontosan. A csoport kutat\u00e1sai az enhancer hat\u00e1smechanizmus meghat\u00e1roz\u00e1s\u00e1ra \u00e9s r\u00e9szletes anal\u00edzis\u00e9re ir\u00e1nyulnak.<\/p>\n<p>In vitro hat\u00e1sm\u00f3d vizsg\u00e1lat:<\/p>\n<p>In vitro vizsg\u00e1lataink sor\u00e1n azt tal\u00e1ltuk, hogy a BPAP, \u00e9s felt\u00e9telez\u00e9s\u00fcnk szerint a tov\u00e1bbi sz\u00e1rmaz\u00e9kok enhancer hat\u00e1sa a TAAR-1 receptoron (trace amine-associated receptor) kifejtett agonista hat\u00e1ssal magyar\u00e1zhat\u00f3. A TAAR szign\u00e1l transzdukci\u00f3s kaszk\u00e1d szab\u00e1lyozza a vezikul\u00e1ris biog\u00e9n amin felszabadul\u00e1st \u00e9s a BPAP katecholamin enhancer hat\u00e1sa az exocit\u00f3zis t\u00f6bb l\u00e9p\u00e9s\u00e9ben is befoly\u00e1solhat\u00f3. Mivel a BPAP katecholamin enhancer hat\u00e1sa ugyancsak kapcsol\u00f3dik a VMAT2 aktivit\u00e1shoz, vizsg\u00e1ljuk a TAAR rendszer \u00e9s a biog\u00e9n aminok vezikul\u00e1ris felv\u00e9tel\u00e9nek k\u00f6lcs\u00f6nhat\u00e1s\u00e1t is.<\/p>\n<p>\u00a0In vivo hat\u00e1sm\u00f3d vizsg\u00e1lat:<\/p>\n<p>Az enhancer hat\u00e1sm\u00f3d vizsg\u00e1lata \u00e9s r\u00e9szletes anal\u00edzise k\u00fcl\u00f6nb\u00f6z\u0151 viselked\u00e9sfarmakol\u00f3giai m\u00f3dszerekkel (tanul\u00e1s, mem\u00f3ria, \u00e1ltal\u00e1nos aktivit\u00e1s, stb.). Mivel az enhancer vegy\u00fcletek jellemz\u0151en a stimulus induk\u00e1lta biog\u00e9n amin felszabadul\u00e1st er\u0151s\u00edtik, az enhancer hat\u00e1s in vivo k\u00f6vet\u00e9se is els\u0151sorban nehez\u00edtett k\u00eds\u00e9rleti k\u00f6r\u00fclm\u00e9nyek k\u00f6z\u00f6tt lehets\u00e9ges.
