Nem teljesen tiszt\u00e1zott azonban, hogy fokozott, tart\u00f3s s\u00f3fogyaszt\u00e1st k\u00f6vet\u0151en milyen molekul\u00e1ris mechanizmusok vezetnek kiterjedt betegs\u00e9gekhez. A k\u00f6zelm\u00falt kutat\u00e1sai h\u00edvt\u00e1k fel a figyelmet arra, hogy a szervezet s\u00f3egyens\u00falya nem kiz\u00e1r\u00f3lag a bevitel \u00e9s a kiv\u00e1laszt\u00e1s f\u00fcggv\u00e9nye. A b\u0151rben, glik\u00f3zaminoglik\u00e1n (GAG) molekul\u00e1kon t\u00f6rt\u00e9n\u0151 Na+ rakt\u00e1roz\u00e1s r\u00f6vid t\u00e1von kedvez\u0151en befoly\u00e1solja a v\u00e9rnyom\u00e1st. Vizsg\u00e1lataink seg\u00edthetik meg\u00e9rteni, hogy e folyamatos ered\u0151jek\u00e9nt mik\u00e9nt alakul \u00e1t Hossz\u00fa t\u00e1von azonban, a b\u0151r alatti s\u00f3rakt\u00e1r lok\u00e1lis stressz r\u00e9v\u00e9n olyan eddig nem, vagy csak r\u00e9szben felt\u00e1rt folyamatokat ind\u00edthatnak el, amelyek a sz\u00f6veti szerkezet \u00e1talakul\u00e1s\u00e1hoz \u00e9s a v\u00e9rnyom\u00e1s v\u00e1ltoz\u00e1s\u00e1hoz vezethetnek.<br \/>\nKutat\u00f3csoportunk \u00e1llat- \u00e9s sejtk\u00eds\u00e9rletes modellek alkalmaz\u00e1s\u00e1val, valamint klasszikus molekul\u00e1ris biol\u00f3giai m\u00f3dszerekkel (qRT-PCR, Western blot, immunhisztok\u00e9mia) vizsg\u00e1lja a s\u00f3terhel\u00e9s sor\u00e1n lerakod\u00f3 Na+ kapcsolat\u00e1t az immunrendszerrel, a lok\u00e1lis RAS, valamint egy\u00e9b fibr\u00f3zist induk\u00e1l\u00f3 rendszerekkel.<br \/>\nVizsg\u00e1lataink seg\u00edthetnek meg\u00e9rteni, hogy e folyamatok ered\u0151jek\u00e9nt mik\u00e9nt alakul \u00e1t a b\u0151r strukt\u00far\u00e1ja, ezen bel\u00fcl a GAG molekul\u00e1k metabolizmusa, a lok\u00e1lis \u00e9rrendszer hemodinamik\u00e1ja, illetve milyen hat\u00e1sokat fejt ki m\u00e1s szervekre.<\/p>\n<p style=\"text-align: justify\"><strong>Le\u00edr\u00e1s laikusok sz\u00e1m\u00e1ra:<\/strong><br \/>\nA magas v\u00e9rnyom\u00e1s \u00e9s sz\u00f6v\u0151dm\u00e9nyei olyan n\u00e9pbetegs\u00e9get jelentenek az iparilag fejlett vil\u00e1gban, amelynek orvosi kezel\u00e9se jelent\u0151s er\u0151forr\u00e1st ig\u00e9nyel, a termel\u00e9sb\u0151l kiesett id\u0151 miatt a t\u00e1rsadalom gazdas\u00e1gi eredm\u00e9nyess\u00e9g\u00e9re is k\u00e1ros, az egy\u00e9n szintj\u00e9n pedig kr\u00f3nikus betegs\u00e9ggel val\u00f3 egy\u00fctt\u00e9l\u00e9st jelenti, amely hossz\u00fat\u00e1von, m\u00e1sodlagosan psych\u00e9s betegs\u00e9gek forr\u00e1s\u00e1v\u00e1 v\u00e1lik. A magas v\u00e9rnyom\u00e1s okozta sz\u00edv- \u00e9s \u00e9rrendszeri betegs\u00e9gek jelentik ma a vezet\u0151 hal\u00e1lokot. Megel\u0151z\u00e9s\u00fckben minden \u00faj r\u00e9szlet \u00e9s eredm\u00e9ny hatv\u00e1nyozott megt\u00e9r\u00fcl\u00e9st jelent. A s\u00f3fogyaszt\u00e1s \u00e9s a magas v\u00e9rnyom\u00e1s, valamint a sz\u00edv \u00e9s \u00e9rrendszeri betegs\u00e9gek k\u00f6z\u00f6tti \u00f6sszef\u00fcgg\u00e9s ismert. Nem teljesen tiszt\u00e1zott azonban, hogy fokozott, tart\u00f3s s\u00f3fogyaszt\u00e1st k\u00f6vet\u0151en milyen molekul\u00e1ris mechanizmusok vezetnek kiterjedt betegs\u00e9gekhez.<\/p>\n<p style=\"text-align: justify\">A kutat\u00e1si projektet arra az \u00faj felismer\u00e9sre alapozzuk, amely szerint a t\u00f6bblet s\u00f3fogyaszt\u00e1s a b\u0151rben s\u00f3rakt\u00e1rakat eredm\u00e9nyez. A b\u0151r alatti s\u00f3rakt\u00e1rak a lok\u00e1lis stressz r\u00e9v\u00e9n olyan eddig nem, vagy csak r\u00e9szben felt\u00e1rt korrekci\u00f3s folyamatokat ind\u00edtanak el, amelyekben aktiv\u00e1l\u00f3dnak az \u00e9rrendszert \u00e9s az egyes szerveket (els\u0151sorban a ves\u00e9t) k\u00e1ros\u00edt\u00f3 feh\u00e9rje-rendszerek is. Ennek a h\u00e1l\u00f3zatnak \u00e9s a benne l\u00e9v\u0151 kulcsmolekul\u00e1knak a felt\u00e1r\u00e1sa, a k\u00f6zt\u00fck l\u00e9v\u0151 \u00f6sszef\u00fcgg\u00e9s le\u00edr\u00e1sa nemcsak \u00faj ismeretekhez vezet, hanem alapul szolg\u00e1lhat \u00faj, eddig m\u00e9g nem alkalmazott gy\u00f3gyszermolekul\u00e1k kifejleszt\u00e9s\u00e9hez is. A kutat\u00e1st d\u00f6nt\u0151en hazai, min\u0151s\u00e9gbiztos\u00edt\u00e1ssal rendelkez\u0151 laborat\u00f3riumban v\u00e9gezz\u00fck, egyes r\u00e9szfeladatokhoz bevonva a legjobb hazai kutat\u00f3kat is. \u00dajdons\u00e1gtartalma miatt jelent\u0151s nemzetk\u00f6zi figyelemre sz\u00e1m\u00edthat.<\/p>\n<p>&nbsp;<\/p>\n<hr \/>\n<div>\u00a0<\/div>\n<div class=\"keretes\"><strong>Supervisor:<\/strong> Dr. Tivadar Tulassay<\/div>\n<p style=\"text-align: justify\"><strong>The effect of salt overload on the immune system and the local renin-angiotensin system of the skin<\/strong><\/p>\n<p style=\"text-align: justify\">Increased salt consumtion is widespread in the developed countries and the relationship between salt consumption and hypertension and cardiovascular diseases is well established. However, the molecular mechanisms leading to extensive diseases following prolonged high salt intake is not clearly defined. Recent research drew attention to the phenomenon, that the salt balance of the body is not exclusively the function of salt intake and excretion.<br \/>\nSalt storage on skin glycosaminoglycan (GAG) molecules of the skin may have beneficial effect on blod pressure in the short term. In the long term however due to local stress subcutaneous salt reservoire may activate hitherto unknown processes which ultimately lead to tissue remodelling and the alteration of blod pressure.<\/p>\n<p style=\"text-align: justify\">Our research group investigates the relationship between Na+ deposition following salt overload, the immune system, the local RAS system(s) and other fibrosis-inducing systems using animal models and classical molecular biological methods (qRT-PCR, western blot, immunohistochemistry). Our research may help us understand how the structure of the skin, especially the metabolism of GAG molecules is altered as a result of the above processes; how the hemodynamcs of the local vasculature changes and what effect they have on other organs.<\/p>\n<p style=\"text-align: justify\">Lay summary:<br \/>\nHypertension and related diseases are among the major public health problems in the developed countries. Their treatment requires extensive financial resources and the lost working hours cause further economic burden. Moreover, in affected individuals the long-term struggle with the disease may result in mental disturbances. Cardiovascular diseases caused by high blood pressure are today\u2019s leading cause of death Their prevention therefore of outmost importance and new discoveries in this field appear to be rewarding. The relationship between salt consumption and hypertension and cardiovascular diseases is well established. However, the molecular mechanisms leading to extensive disease following prolonged high salt intake is not clearly defined.<br \/>\nOur research project is prompted by recent findings that excessive salt consumption and salt load is buffered by the generation of osmotically inactive salt reservoir in the subcutaneous intestitium. Due to the local stress this reservoire may initiate protective processes, and on the other hand it may activate protein networks involved in cardiovascular and other organ damages (the kidney in particular). The identification of the key molecules of this system and the discovery of their interactions would help us to better understand the blood pressure regulation and to develop novel, more targeted drug therapies. The research will be carried out in our accredited laboratory with the participation of dedicated scientists.
